Will a 15x7 wheel with a 3.75 backspace work on the rear of a 66 without clearence problems?

yes, the wheel will, but what kind of tire are you mounting is the question. That wheel doesn't have much backspacing so the wheel will tend to stick out of the quarter panel.

I have 215's on it now and wanted to use the existing tires. Should I go with a larger backspacing?
